00:00:16.03 Good morning. My name is Julie Huber,
00:00:18.09 and I'm a microbial oceanographer at the Marine Biological Laboratory
00:00:21.09 in Woods Hole, Massachusetts.
00:00:23.03 Today, I want to tell you about some of the work I do studying microbes in the deep sea,
00:00:26.28 why I think it's important, and what it's like to be a field-going, ocean-going scientist.
00:00:31.01 I wear a lot of different hats as a scientist,
00:00:34.18 but I think this cartoon illustrates the core of what I do,
00:00:37.19 which is I use cutting-edge technology to go to strange places
00:00:41.13 at the bottom of the ocean
00:00:42.09 to look for microbial life.
00:00:44.05 In fact, the surface of our planet is covered in water.
00:00:48.23 About 70% of Earth is covered in water,
00:00:51.05 but beneath the surface, some pretty amazing things are going on.
00:00:54.08 This is a topographic map, where we've taken away all the water
00:00:57.20 so you can see the depth or the height of everything on the planet,
00:01:00.22 and particularly, in the blue parts, where our oceans are,
00:01:03.20 you can see there's a lot of interesting relief.
00:01:05.29 In particular, things like this, where you can see mountain ranges beneath the sea floor.
00:01:10.20 This is the world's longest mountain range. It's about 55,000 kilometers long,
00:01:15.17 and it represents places on the sea floor where tectonic plates are moving apart.
00:01:19.24 When tectonic plates move apart, volcanism results,
00:01:23.20 and that's illustrated in this slide.
00:01:25.15 When tectonic plates either spread apart, you get things like mid-ocean ridges.
00:01:29.16 When plates are subducting beneath one another, you get volcanic arcs.
00:01:33.22 In fact, volcanism is pervasive underneath the ocean,
00:01:37.11 and about 70% of the volcanism on our planet occurs within the water.
00:01:41.17 However, until recently, we didn't know much about it
00:01:44.29 because we didn't have the ability to study it and observe it.
00:01:48.00 Here's one example, however, when we were able to.
00:01:51.15 This is a deep-water eruption, occurring at about 4,000 feet depth
00:01:55.05 from an expedition I participated in near Samoa in 2009.
00:01:59.06 Until we took this footage, just a few years ago,
00:02:02.11 no one had ever witnessed this fundamental Earth process occurring,
00:02:05.20 despite the fact that it's probably been going on
00:02:08.05 since our planet formed about 4.5 billion years ago.
00:02:11.02 So, we had little understanding of how new ocean crust formed,
00:02:14.20 what types of volatiles were being pushed out of volcanoes,
00:02:17.13 and how and if life could survive in this type of environment.
00:02:21.05 So, with this footage, and with these types of expeditions,
00:02:24.11 we're able to make amazing strides in our understanding in both life and Earth processes.
00:02:29.12 This volcano is emitting all sorts of volcanic gases,
00:02:34.24 which serve as very good food for microbial life.
00:02:37.16 That includes things like hydrogen sulfide
00:02:39.10 that microbes, bacteria, and archaea can exploit.
00:02:43.11 So, at deep sea hydrothermal vents, microbes are forming the base of the food chain.
00:02:48.12 This is footage from the Galapagos ridge.
00:02:50.18 That's where hydrothermal vents were first discovered in 1977.
00:02:54.12 These are giants tube worms, and the only reason
00:02:57.06 these tube worms are able to survive in this environment
00:02:59.23 is because their gut is packed full of bacteria that are oxidizing hydrogen sulfide
00:03:04.19 from the vent fluid and creating organic carbon that the worms use to live.
00:03:08.25 This is in contrast to photosynthesis, where you have light providing the main energy
00:03:14.03 source and supporting the base of our food chain on land.
00:03:17.17 Until the discovery of these hydrothermal systems in 1977,
00:03:21.24 no one even knew chemosynthetic life was possible.
00:03:24.27 So, the discovery of hydrothermal vents completely changed
00:03:27.11 the way view biology on our planet.
00:03:29.19 And it's a very good example of how not everything is hypothesis driven.
00:03:34.00 This was not explained, or not hypothesized,
00:03:37.05 and was a complete surprise to all of biology.
00:03:39.29 So, it isn't just hydrothermal systems that are dominated by microbes.
00:03:44.04 In fact, we live on planet microbe.
00:03:46.04 You might feel like it's a eukaryotic world because
00:03:48.15 we're surrounded by trees, animals, and humans,
00:03:50.29 but in fact, microbes mediate all the major biogeochemical cycles on our planet.
00:03:55.04 Carbon, oxygen, nitrogen... these are the things that make Earth habitable for higher life.
00:04:00.18 In addition, microbes were the only form of biological life
00:04:04.28 on our planet for about 3.5 billion years.
00:04:07.27 Remember, our planet is about 4.5 billion years old.
00:04:10.18 So, they're the dominant form of life for most of Earth's history.
00:04:13.20 That's in contrast to modern humans.
00:04:15.13 We've only been around for a few hundred thousand years.
00:04:17.25 So, in that time, microbial communities have had time to evolve,
00:04:21.28 and they occupy nearly every corner of our planet, even the human body.
00:04:26.23 And if we look at the universal tree of life,
00:04:29.21 with all the known realms of life put on to the same tree,
00:04:34.07 we can see that most of life on this tree is, in fact, microbial.
00:04:38.10 They belong to two different domains: the bacteria and the archaea.
00:04:42.03 These are single-celled organisms. They divide by binary fission.
00:04:45.17 They don't have any membrane-bound organelles, but you can see
00:04:48.13 they occupy a huge amount of diversity
00:04:50.15 because they've had so long to evolve on our planet.
00:04:53.08 And just in contrast, we are right here, at the very tip of the tree.
00:04:57.09 So, you can see that most of the life on our planet is microbial.
00:05:00.21 So, all of the work in my lab, here at the Marine Biological Laboratory,
00:05:04.27 focuses on understanding these microbes.
00:05:07.12 And we use a variety of tools to do that.
00:05:10.00 We want to understand what they eat, how quickly they grow,
00:05:12.25 what are their genes doing? How do they evolve?
00:05:15.17 How are they interacting with and impacting the local environment?
00:05:18.10 So, we use molecular tools that involve looking at DNA and RNA from these organisms.
00:05:23.24 We try to culture them in the laboratory, which is shown down here.
00:05:27.04 And we then use microscopy and geochemical tools to study them.
00:05:31.12 I'm particularly interested in microbes that are specifically adapted
00:05:37.03 to the volcanic habitat. So, this includes organisms that don't use oxygen
00:05:41.06 and are using these volcanic gases to make a living.
00:05:44.08 So, this is one example. These are methanogens.
00:05:46.21 These belong to the domain Archaea.
00:05:48.18 Methanogens use carbon dioxide and hydrogen to generate methane,
00:05:52.07 and they're very successful in these ecosystems.
00:05:55.16 Another group that belongs to both bacteria and archaea are sulfur reducers.
00:06:00.12 Sulfur reducers again use sulfur coming out of the vent system,
00:06:03.19 combine it with hydrogen, to produce hydrogen sulfide.
00:06:06.16 So, both of these organisms... neither of them use oxygen,
00:06:10.12 and the discovery of organisms surviving like this, in deep sea systems,
00:06:14.17 often at high temperature,
00:06:16.07 led a number of people to hypothesize that maybe
00:06:18.21 they are remnants of Earth's earliest life forms.
00:06:21.14 So, remember that I told you, hydrothermal systems are very ancient on our planet,
00:06:25.20 and so if we've had volcanism going on for a very long time,
00:06:28.24 these types of organisms are potentially some of the earliest life forms
00:06:32.22 that we've had on our planet.
00:06:34.14 In addition, the discovery of hydrothermal systems
00:06:37.10 opened a whole new way of looking at life beyond our own planet.
00:06:41.00 So, the field of astrobiology involves looking at the
00:06:43.22 origin and evolution of life beyond Earth.
00:06:46.02 And this is Mars and Europa shown here,
00:06:48.21 two very promising candidates for either current or past life.
00:06:52.19 For example, we know that there's a history of volcanism in the past on Mars,
00:06:56.29 and we believe that there's a salty ocean beneath the icy shell of Europa,
00:07:01.01 one of Jupiter's moons.
00:07:02.09 Could there be hydrothermal systems beneath that icy shell?
00:07:05.27 Could there be life beneath that icy shell?
00:07:08.18 So, this whole combination of interesting reasons to study hydrothermal systems:
00:07:12.23 the fact that they're ancient, the fact that we can understand
00:07:15.14 very fundamental Earth processes going on beneath the ocean's surface,
00:07:18.22 and the fact that we could potentially learn more
00:07:20.29 about the origin and evolution of life beyond Earth,
00:07:23.26 really led me down this road as a young undergraduate student.
00:07:27.19 I went off to college and thought, like many kids,
00:07:30.06 I was going to be the next Jacques Cousteau.
00:07:31.29 I was going to study dolphins and coral reefs and sharks.
00:07:35.04 But I became really engaged by this possibility of studying microbial life,
00:07:39.03 not only as powering our own planet, but planets beyond our own.
00:07:42.25 So, most of the work in my laboratory takes place... it looks very normal,
00:07:47.23 like what you're probably used to seeing in any biological laboratory.
00:07:51.22 We have PCR machines, we have culturing, we have all sorts of basic laboratory things.
00:07:56.28 But to have anything to work with, we have to go to a very different laboratory.
00:08:01.05 In this case, it's the bottom of the ocean.
00:08:03.23 So, this is a shot of me diving in the man submersible, Alvin.
00:08:07.00 I'm sitting at about 4,000 feet beneath the surface,
00:08:10.01 collecting microbes for our study.
00:08:12.03 So, in this case, our laboratory becomes a ship,
00:08:15.03 and our field site, the bottom of the ocean.
00:08:16.21 This is the Atlantis, which is the mothership for Alvin,
00:08:21.00 and we usually go out to sea for 2-4 weeks at a time to collect our samples
00:08:25.15 and then eventually bring them back to the laboratory for further study.
00:08:28.26 So, I already mentioned Alvin, which is the deep sea submersible that we use
00:08:33.22 for some of our collections, but often that isn't practical or safe.
00:08:38.24 For example, that exploding volcano that I showed you at the very beginning.
00:08:41.24 That's not the type of place that you would want to bring
00:08:43.24 humans at the bottom of the ocean.
00:08:45.10 So, in that case, we use remotely operated vehicles.
00:08:48.01 This one is Jason. It's operated out of the Woods Hole Oceanographic Institution,
00:08:52.15 and it's tethered up to the ship by a power cable. There's no people on board the vehicle,
00:08:58.05 but instead, we are all back up on the ship, in a dark room,
00:09:01.12 with about 14 different monitors
00:09:03.03 that are showing us what's happening on the sea floor.
00:09:06.08 And we work with a team of pilots and engineers that run the ROV,
00:09:09.27 and they help us collect our samples,
00:09:11.17 make our maps, and make good decisions about how
00:09:14.05 best to do our science on the sea floor.
00:09:16.04 We also use something called autonomous underwater vehicles.
00:09:20.12 This one is called Nerius,
00:09:22.02 and this is a vehicle that allows us to completely set the vehicle free from the ship,
00:09:27.05 and it can run a map, for example. It has sensors on it.
00:09:30.01 And then we can use the ship to go do other things.
00:09:32.18 And, the final thing we have to do, which is perhaps
00:09:35.21 one of the most challenging parts is design very specific samplers
00:09:38.18 for the types of samples that we're after.
00:09:41.14 And, we're always sort of constrained by the vehicle we're using,
00:09:44.28 the study site, the depth we're operating at,
00:09:47.13 and often I walk off a ship and just think,
00:09:51.02 "Wow, that was 50% of my job, simply getting those samples,"
00:09:54.28 and then I have a whole layer of another work to happen in the laboratory.
00:09:58.14 So, working at sea is a pretty unique place to do your job.
00:10:03.11 It's very challenging. It's very humbling sometimes,
00:10:07.17 particularly to be sitting in the middle of the ocean
00:10:09.13 and not really see anything for days at a time.
00:10:12.04 We run a lot of different safety drills because, of course,
00:10:16.22 we are on our own, in most cases.
00:10:18.15 So, this is a shot of me off the coast of Hawaii, running through our weekly safety drill
00:10:23.09 to make sure if I had to abandon ship that I would know how to do that.
00:10:26.09 And another particular challenge is that we're working on a moving platform,
00:10:31.09 and so, we're dealing with chemical... needles in this example,
00:10:35.01 on a moving platform, and sometimes just keeping your balance,
00:10:38.03 keeping your breakfast down...
00:10:39.22 Things can be particularly challenging, just doing basic lab work at sea,
00:10:43.28 which is why when we're at sea, we mostly just focus on getting the samples,
00:10:47.19 and we do most of the analysis when we get back home.
00:10:49.27 We figure a lot of things out.
00:10:52.19 And, I like to say that being at sea is sort of like being at camp in some ways,
00:10:57.20 except you're with grown-ups.
00:10:58.20 So, here I am making coffee. We play ping-pong. We do laundry.
00:11:03.25 There's a gym. We eat all our meals together.
00:11:06.21 So, it's a very ... bunch of grown-ups on a 280 foot vessel,
00:11:10.16 hanging out for a few weeks, working a lot,
00:11:12.10 but we also manage to find some creature comforts.
00:11:14.22 And all of this is really for the privilege of doing things like diving
00:11:19.05 to the bottom of the ocean and observing things
00:11:21.07 that very few people get to observe in first person.
00:11:23.20 This is me diving ... going into Alvin at sunrise one morning
00:11:27.14 before heading to the bottom of the Pacific Ocean.
00:11:29.15 But, perhaps, my favorite thing about going to sea
00:11:32.12 is the fact that I don't work in isolation.
00:11:34.23 So, a lot of people think about a scientist working in their laboratory
00:11:37.17 late at night, by themselves, making discoveries.
00:11:40.07 In fact, some of the most remarkable discoveries we've made have been as a team.
00:11:44.02 About 50 of us strong, including scientists, crew members, from everybody --
00:11:48.14 from the guys working in the engine room to the people washing dishes,
00:11:51.14 to all of us operating the vehicles and doing the science.
00:11:54.18 And I think it was this interdisciplinary nature of oceanography
00:11:57.29 that drew me to it. I'm not a very lonely person, and I like working with other people
00:12:03.14 and to be at sea with a bunch of people who have left their families,
00:12:06.21 who have left their desk job to come out
00:12:08.29 and really focus on a particular system or question for me
00:12:12.13 is incredibly exciting.
00:12:14.13 And there's still a lot to do.
00:12:16.23 I showed you that map of the ocean. It's a big ocean.
00:12:20.01 We have very few studies of deep oceanic volcanic systems.
00:12:24.13 And so, I figure, as long as volcanoes continue to thrive on our planet
00:12:28.20 and life finds a way to study them,
00:12:30.09 I will have a job.
00:12:31.28 And, I think I still have plenty to do to drive many careers ahead of me.
00:12:36.04 So, I hope I've given you a taste of why volcanic systems are important,
00:12:39.29 why the life there is so interesting,
00:12:41.22 and a little bit of what it's like to be a sea-going scientist.
