DNA Replication: Understand How DNA is Replicated

00:00:06.20 Hello, I'm Bruce Alberts and I'm going to talk about something I spent 30 years on,
00:00:13.10 trying to understand how DNA is replicated, a very fundamental process in all living systems.
00:00:22.11 I'm going to give you a little history because I think that outlining a pathway to discovery
00:00:30.00 where we were lost in the woods, not knowing where we were going
00:00:35.04 and misunderstanding things can help to make sense of what we now know.
00:00:39.00 It also, trying to convey a sense of how science works, in this short talk.
00:00:44.07 So, this is a picture taken when I was in high school, 1953,
00:00:50.24 a major breakthrough in our understanding of life
00:00:53.08 happened when Watson and Crick correctly predicted the structure of DNA double helix
00:01:01.12 and as soon as they got that structure, our understanding of living things,
00:01:08.15 and specifically of heredity, changed dramatically.
00:01:11.14 I'd like to point out that at the time, Watson was only 25 years old.
00:01:16.07 I'm a big supporter of young scientists,
00:01:18.17 most creativity on the part of science happens when they're much younger than I am.
00:01:27.03 This is the famous paper, first paper published on the double helix
00:01:33.02 and it has this famous sentence, outlined in yellow here:
00:01:37.18 "It has not escaped our notice that," basically our model suggests
00:01:44.16 how genetic information is transferred between one generation and the next.
00:01:49.18 They were correct.
00:01:51.22 Now, the point is, they subsequently published a short paper in the same year
00:02:00.07 which basically outlined this scheme,
00:02:04.26 called semi-conservative DNA replication,
00:02:08.06 where each old strand of the DNA double helix templates the formation of a new one
00:02:13.28 using the four base pairs that the observed in the DNA double helix.
00:02:20.05 But they had, it's important to note, that they had no idea how this actually would happen.
00:02:26.10 This, they understood in principle how it could happen
00:02:30.16 and the next major breakthrough came only a few years later,
00:02:35.13 a biochemist, Arthur Kornberg, who also earned the Nobel Prize,
00:02:40.20 as did Watson and Crick, discovered the enzyme DNA polymerase.
00:02:44.20 This enzyme was thought to be very remarkable at the time.
00:02:51.14 It will add a new nucleotide to the end of one DNA strand, called the primer strand,
00:02:57.05 only if that strand is paired to a complementary strand, that's to serve as its template
00:03:01.26 and there are two problems, basically I'll describe with their finding
00:03:07.29 as far as really understanding how it worked.
00:03:12.15 Because they thought the template strand must be single stranded
00:03:15.18 with its bases exposed, and second of all, that the DNA polymerase enzyme
00:03:21.00 moves in only one chemical direction, that is growing the 3 prime (3') end of a new DNA strand.
00:03:28.17 So, here's a little schematic view, primer gets added onto one nucleotide at a time,
00:03:33.29 which nucleotide gets added depends on what base is on the template strand at that point
00:03:40.22 and the 3' end of the primer extends one nucleotide at a time.
00:03:48.19 So, this is a more detailed view, showing some of the chemistry,
00:03:54.17 we actually, as discovered by Kornberg, the source for the energy of polymerization
00:04:00.24 is an incoming deoxyribonucleoside triphosphate
00:04:05.08 and the two phosphates that are marked in red get split off every time this single unit is added to the 3' end.
00:04:16.01 And again, as you can better see here,
00:04:19.17 which base is added depends on specific base pairing, here C with a G,
00:04:24.15 the G is on the template, C is coming in as a new monomer.
00:04:30.09 So, I started working on DNA replication, way back then, I was a beginning graduate student
00:04:38.19 and I was fascinated by trying to solve the problem of how DNA might replicate,
00:04:45.05 what was the machinery that would carry out the process that wasn't predicted
00:04:50.15 and one of my professors, a famous professor,
00:04:53.09 talked about, I still remember him giving this lecture about how it might work
00:04:57.24 and here's one the models that was suggested.
00:05:01.16 Since the DNA polymerase only goes in one direction,
00:05:04.25 maybe replication only starts at one end of the chromosome
00:05:10.27 and opposite ends get synthesized going along this mechanism diagrammed here
00:05:20.08 and I just want to show you, this would create a lot of single stranded DNA.
00:05:24.12 Because this strand opposite the template strand would be released into a DNA single strand
00:05:32.08 and so, if you looked at a replicating chromosome, you should see a lot of single-stranded DNA.
00:05:39.20 Well, just one year later, a man by the name of John Cairns did a famous experiment
00:05:46.23 where he actually did electron microscopy of what you see when DNA is replicated.
00:05:52.00 He discovered the replication fork. That is there isn't any of this large amounts of single-stranded DNA.
00:05:59.21 Instead, DNA is replicated on both sides, both strands of DNA double helix roughly at the same time.
00:06:09.00 And so at a replication fork, the two daughter DNA strands appear to grow at opposite ends,
00:06:16.16 and yet we know that DNA polymerase only goes one way. How is this possible?
00:06:22.05 This was the mystery that faced me when I started work in the lab as a graduate student.
00:06:30.07 And this was the way people drew it, actually,
00:06:35.13 same professor is famous for drawing a fig leaf a little later over the replication fork.
00:06:41.12 Saying we had no idea what happened at that point.
00:06:46.26 So, 1961, when I started my work as a graduate student
00:06:52.03 and when I did this, I was given the freedom to choose anything I wanted to work on,
00:06:57.00 that was wonderful, and I wanted to solve the mystery of how DNA replicated.
00:07:01.13 We assumed, everybody assumed that a single enzyme, DNA polymerase,
00:07:06.17 alone replicated DNA. But how was this possible?
00:07:09.22 Two problems of course that DNA polymerase works only on single stranded DNA
00:07:15.18 and yet DNA is a double helix inside the cell, as Watson and Crick showed,
00:07:19.17 and secondly, the two DNA strands, as Watson and Crick showed
00:07:23.21 went in opposite directions, they were antiparallel.
00:07:26.08 And yet the polymerase only goes one way.
00:07:29.26 Well, this led to very unsuccessful career as a graduate student
00:07:38.12 because I had been inspired by Watson and Crick to believe
00:07:42.27 that theoretical biology could predict things
00:07:46.05 and I think not only me, but many graduate students were predicting things and trying to test their ideas,
00:07:52.04 we're more sophisticated now, so I predicted how DNA polymerase might be able to start DNA synthesis on a chromosome,
00:08:00.17 I actually used it as a term paper in one of my graduate courses and I got an A+,
00:08:06.17 so, oh my goodness, this must be right.
00:08:09.09 So, I then spent years doing experiments to see if my theory was right.
00:08:14.01 Of course, my theory wasn't right, and as you'll see, the whole hypothesis was wrong,
00:08:19.00 that one enzyme could replicate DNA.
00:08:21.16 And I had a very unsuccessful, difficult time as a graduate student, from which I learned a lot,
00:08:26.22 you could hear about that if you go through the short talk,
00:08:31.11 I've given on iBio as well, it's also here in a section about lessons learned from failure
00:08:39.08 and I've had many failures, that's one of them I explained how failure's important for a career.
00:08:46.15 You have to learn from your failures, that's what makes you better the next time.
00:08:51.03 But then I finally got my PhD and I moved to Geneva, Switzerland for a year as a post-doctoral fellow.
00:08:56.13 I immediately discovered, to my great surprise and someone consternation
00:09:02.00 that if I had been reading the scientific literature I would have known better
00:09:06.10 because work done in bacteriophage T4, a wonderful model system for early stages of molecular biology,
00:09:15.14 it's a large, double-stranded chromosome virus, infects E. coli and it makes all its own replication machinery
00:09:24.00 and it had about a hundred genes discovered by genetics and published in 1963,
00:09:28.05 in the middle of my thesis and the fact is that there were at least 7 genes in this virus
00:09:37.02 that were absolutely required for DNA replication of this virus,
00:09:40.05 but the geneticists who did this work had no idea what these genes did.
00:09:46.04 They gave them numbers, 32, 41, 43, 44, 45, 61 and 62.
00:09:52.23 And a little later, one of these genes, the gene 43 protein,
00:09:57.25 had been shown to produce T4 bacteriophage DNA polymerase,
00:10:01.07 so that we understood, that made sense.
00:10:03.21 But the question was, why are these six additional proteins needed
00:10:06.29 for any replication of the T4 chromosome when the virus infects E. coli.
00:10:12.07 And this meant that I and others had been completely naive
00:10:16.25 in thinking that only a single protein was needed.
00:10:19.18 These sub-proteins must be needed and therefore replication must be much more complicated than anyone had imagined.
00:10:27.14 So then I was getting ready to start my own lab and I needed a strategy
00:10:33.11 and how was I going to, every young, starting scientist really should have the opportunity to start a new problem from scratch,
00:10:42.11 that's the way we get innovation,
00:10:46.18 and again, I was still devoted to solving this problem that I had failed to solve
00:10:52.27 and I had a new strategy, a completely different one.
00:10:56.18 I decided I would make a column out of DNA
00:11:00.13 and looked at all the proteins that bind to DNA
00:11:03.03 in that column, as a biochemist, break open the cell, pass the crude extract over the column,
00:11:10.01 and some of the proteins, a small fraction of them,
00:11:11.29 will bind to the DNA column, they could be then removed from the DNA column
00:11:16.23 by raising the salt concentration and I found that many proteins in a T4 infected cell,
00:11:22.07 made by the T4 bacteriophage genes, bound to this column.
00:11:26.00 Then the question was, would one of these proteins be missing if a mutant virus is used
00:11:30.20 instead of the normal virus from the infection.
00:11:33.10 So, then I could find a protein that the genetics said was need for the very mysterious machinery that replicated DNA.
00:11:43.09 Here's a slide taken from an experiment we published in Cold Spring Harbor Symposium, 1968,
00:11:51.10 the first real discovery from my independent lab,
00:11:54.17 at that time at Princeton. We had done that experiment I just described
00:12:01.17 and found that when you infected a cell with a gene 32 mutant,
00:12:09.18 a major band that was present in all the other infections was missing.
00:12:14.18 You know, pretty obvious, and because it was missing only when you had a gene 32 mutant,
00:12:22.15 we concluded that that must be the product of gene 32,
00:12:26.20 called the 32 protein and that therefore, it must also be required for DNA synthesis.
00:12:34.14 But of course, we had no idea what it did, so we had to do biochemistry
00:12:38.24 and basically, what we discovered is drawn in this little schematic.
00:12:46.17 The 32 protein binds tightly and cooperatively to single-stranded DNA,
00:12:52.12 forming long clusters on the DNA. In so doing, it straightens out the DNA molecule
00:12:59.18 because DNA, single-stranded DNA, single stranded RNA, would otherwise be folded into little knots
00:13:08.06 because these little hairpin helices that can form with imperfect base pairing
00:13:14.18 and this open form of DNA is the kind of DNA that DNA polymerase can more effectively use as a template.
00:13:26.10 So, here's one of our great experiments, you know, seeing is believing,
00:13:33.13 I collaborate with electron microscopist Hajo Delius at Cold Spring Harbor
00:13:36.23 and here's what we found.
00:13:39.23 When we took circular DNA single strands, those are actually the viral DNA from a small virus,
00:13:48.24 but it's completely circular and single-stranded,
00:13:52.21 and without 32 protein, it's this little ball,
00:13:56.16 you see an example with that arrow, without 32 protein, it's this little ball,
00:13:59.25 all tangled up in a little knot, when you add 32 protein, the whole molecule's extended
00:14:05.09 in a beautiful way, in the form that turns out leaves the DNA bases exposed
00:14:13.25 so that DNA polymerase can still see them.
00:14:16.24 So, if we now go to the next slide, this is what we conclude
00:14:22.18 after doing a number of experiments,
00:14:25.11 that the form in which the 32 protein holds the DNA, it's a much better template for DNA polymerase,
00:14:33.05 allowing it to go much more rapidly than it would go otherwise,
00:14:37.23 over this structured single-stranded DNA which would, the form of DNA inside the cell
00:14:46.05 without the 32 protein.
00:14:48.06 Now, there were 5 mysterious proteins left.
00:14:50.28 Of course, we looked for some of the other weak bands on our columns, we didn't see them,
00:14:57.07 so we tried a different approach called an in vitro complementation assay.
00:15:02.02 It was a very simple idea, if you break open the infected cell
00:15:07.15 and add precursors to DNA, we see that some of that precursor nucleotides are incorporated into DNA.
00:15:17.25 A very little amount, it's a very bad reaction because everything's being degraded as it's being made
00:15:25.03 and everything's mixed together inappropriately,
00:15:27.20 but with that assay, we asked, could we find the stimulation of DNA synthesis in this crude mixture
00:15:32.29 that depended on adding one of the five missing proteins
00:15:36.25 and fortunately for us, it worked for all the missing proteins.
00:15:41.23 And this is the kind of data we got, basically, if you added any extract of an infected cell
00:15:55.20 to a crude lysate, made with a mutant, say in this case, it was 62 mutant,
00:16:01.02 you could get a little stimulation very rapidly, stopped this lousy DNA synthesis,
00:16:10.25 two-fold stimulation in the amount of DNA made,
00:16:13.10 if you added the original extract of course, or uninfected cells, nothing happened,
00:16:18.03 so we had an assay and soon as a biochemist has an assay,
00:16:21.19 you can purify anything, by following it through a series of chromatography steps.
00:16:28.07 This work was mostly done by my graduate students at Princeton
00:16:31.24 and finally, after years of collecting each of these proteins,
00:16:37.07 every time we got a new one, we added it to DNA polymerase and see if it would replicate.
00:16:42.05 The answer was no, but when we had all 7 and only when we had mixed all 7,
00:16:47.05 suddenly, we had this magic day, which remember, I'd been working on this since 1960,
00:16:52.17 very unsuccessfully, now we could do what I could never do before,
00:16:58.08 replicate a double-helical DNA template. If we left out any one of those seven proteins,
00:17:05.05 it didn't work. Now, important point here is that this kind of in vitro biochemistry with purified proteins
00:17:14.05 is essential if we're going to understand how life works because only then can you manipulate the system
00:17:20.28 to actually see what actually is going on.
00:17:23.21 And it took us another 6 years then to figure out why each of the proteins was needed in this reaction.
00:17:28.27 I'm not going to go through all the details by any means,
00:17:32.01 I'll just give you one example that's simple.
00:17:35.07 The gene 45 protein turned out to form a sliding clamp for the DNA polymerase
00:17:39.16 which allowed the polymerase to move for many thousands of nucleotides,
00:17:42.28 hundreds of thousands, without dissociating.
00:17:45.12 This is how we viewed DNA replication in the old days.
00:17:50.15 This is how we viewed life in the old days; basically everything was a random collision.
00:17:54.20 And so here's DNA polymerase, coming in, adding one nucleotide, falling off,
00:18:00.04 a second DNA polymerase, because collisions are very rapid in the cytosol,
00:18:04.18 in chemistry, we knew that from chemistry, so the second DNA polymerase comes in,
00:18:09.23 it adds another one, then it falls off, another one comes in.
00:18:12.21 Well, it turns out that kind of mechanism for life in general
00:18:16.18 is so inefficient relative to what actually happens that it's only rarely used.
00:18:22.25 Usually things are much more organized, as I'll explain.
00:18:25.17 Turned out that the 45 protein produced a sliding clamp for the DNA polymerase
00:18:31.24 that we can show biochemically enabled the DNA polymerase to go for hundreds of thousands of nucleotides,
00:18:36.26 we tried to figure out why that was so and I had a graduate student who worked very hard on this,
00:18:43.03 but we missed the fact that, as first discovered in the E. coli system by Mike O'Donnell,
00:18:50.12 the reason why it worked like this is it was a ring,
00:18:54.17 it formed a ring around the DNA that created a tether for the DNA polymerase
00:18:59.05 and the DNA polymerase is kept associated with the same DNA molecule because it's pulling this ring along as it moves.
00:19:07.13 And this is a structure that was solved originally for the E. coli protein.
00:19:15.00 I've pointed out on several slides that there are human versions of all these proteins.
00:19:22.23 In this case, the human protein is called PCNA, it looks the same.
00:19:27.09 A ring, pulled along by DNA polymerase.
00:19:31.04 So, I skipped an important point here because I was going about, talking about what I was doing,
00:19:40.15 but early in our work, by 1970 or so, we knew from the work of a lab in Japan,
00:19:48.11 two Okazaki, husband and wife, and their co-workers,
00:19:55.14 that when you looked at the newly synthesized DNA inside of a cell,
00:20:00.24 actually they used radioactive post-labeling to see what the new DNA was like,
00:20:06.14 the DNA, half the DNA, that made on the so-called lagging strand,
00:20:12.26 the strand that was so mysterious how you could move at a replication fork,
00:20:16.27 that DNA was made backwards, again, it's made by the same DNA polymerase,
00:20:22.00 which can now go backwards, because that's the direction it goes,
00:20:25.13 but it's made as a short series of fragments, which are subsequently run into each other
00:20:32.01 and sealed together to create a continuous strand
00:20:35.22 on the lagging strand of the DNA replication fork,
00:20:41.28 just like the DNA on the leading strand is continuous because it's made continuous.
00:20:48.05 Now this is a mechanism that was completely unpredicted.
00:20:52.13 I think if anybody had predicted this mechanism in 1960,
00:20:58.20 they would have said you're crazy, because this is too sloppy, it would create a mess,
00:21:05.05 all these fragments would get mixed up and you would have all kinds of errors.
00:21:08.17 But in fact, this is the way life works.
00:21:11.29 More specifically, each Okazaki fragment was made by the same DNA polymerase on the leading strand,
00:21:21.15 that DNA polymerase needs a special primer to get started
00:21:25.25 and that primer must be made very frequently on the lagging strand,
00:21:31.22 to start each of the Okazaki fragments
00:21:33.26 every hundred or thousand nucleotides depending on the system
00:21:37.17 and this requires a separate enzyme. It turned out to be one of the seven proteins
00:21:43.21 that was so mysterious in our system. This enzyme is called DNA primase,
00:21:48.22 it makes a short fragment that DNA polymerase then takes over.
00:21:53.25 Very importantly, because you don't want RNA in your final DNA product,
00:21:59.08 that RNA is recognized and specifically erased before all these fragments are sealed together
00:22:05.08 in the end to make a continuous leading strand.
00:22:08.20 Now, in retrospect, we could understand why DNA replication is so complicated.
00:22:16.03 We never predicted it. It's because of the need for enormous accuracy.
00:22:20.11 DNA polymerase turns out to have two active sites,
00:22:26.08 one for polymerization and one for proofreading.
00:22:30.11 Why is proofreading needed?
00:22:33.02 Proofreading is needed because although we might imagine
00:22:36.16 that the base-pairing that Watson and Crick discovered would be perfect,
00:22:41.18 A could only pair with T and C could only pair with G,
00:22:46.01 this violates all the rules of chemistry and life must exist with chemistry
00:22:50.22 as it is in the real world and it turns out that you can't have perfection in chemistry
00:22:58.12 and bases have rare tautomeric forms which are of higher energy,
00:23:03.21 but nevertheless exist in one part in ten to fourth or so,
00:23:07.00 one part in 10,000, and in their incorrect, minor, tautomeric form, they can pair incorrectly
00:23:16.02 and for example, a C could pair with an A, be added by DNA polymerase as it's moving.
00:23:22.07 DNA polymerase evolved so it could recognize that mistake
00:23:26.17 because, when it, before it will add onto the previous place where it just added,
00:23:33.23 it makes sure that that 3' end is base paired and so, as indicated in this diagram,
00:23:40.22 it can recognize that something's wrong, back up, chop off the mistake it made and go forward again.
00:23:47.16 And this, it turns out, to increase the accuracy by something like a factor of a hundred
00:23:54.09 over what would happen without self-correcting polymerase.
00:23:57.01 Anyway, to make a long story short, I'm not explaining this really,
00:24:02.03 only the enzyme that goes in this direction can do that effectively.
00:24:07.07 We understand that and that's why we don't have DNA polymerase going continuously on the lagging strand.
00:24:13.10 It's also why DNA polymerase must have a primer to start,
00:24:18.11 it can't start a new chain by itself because it looks for a perfect pairing before it can start
00:24:23.16 and thus, also explains why an erasable RNA primer makes so much sense
00:24:30.08 and why you need a DNA primase enzyme.
00:24:34.09 Well, the net result of all this, plus things I can't talk about,
00:24:39.22 is DNA is replicated with only about one mistake per billion base pairs that are copied.
00:24:45.25 One part in 10 to the 9th.
00:24:48.19 This in incredible. Humans can't do anything like this.
00:24:53.22 And we know that without such accuracy, a complex organism like a human could not exist.
00:24:59.21 We'd have too many mutations, one of the things that would happen is
00:25:04.18 that every embryo would get cancer and be destroyed.
00:25:09.11 Many other things would go wrong.
00:25:10.28 And so what we could imagine is that as cells evolve to more and more complex forms,
00:25:18.07 all these mechanisms got added and somehow the original mechanism
00:25:23.26 by which DNA replicated obviously couldn't have been this complicated,
00:25:27.21 but after all these many, many rounds of evolution, billions of years of natural selection,
00:25:34.12 we ended up with the mechanism that I was fortunate to be involved in helping to discover,
00:25:41.24 which was of course a real privilege and a great pleasure to solve this mystery.
00:25:48.26 So, here's the DNA replication fork with its proteins on it.
00:25:52.18 I didn't even talk about some of these.
00:25:55.15 Let me, I talked about most of them.
00:25:57.16 The two green molecules are the DNA polymerase, one on the leading strand,
00:26:02.03 and one on the lagging strand, same, in our system it's exactly the same DNA polymerase molecule,
00:26:07.20 the red ring behind it is the sliding clamp that keeps the polymerase from dissociating from the DNA,
00:26:16.10 the DNA polymerase is tethered by this clamp to the DNA.
00:26:21.00 On the leading strand, this is all very simple, it just keeps going.
00:26:25.05 On the lagging strand, it's very complex.
00:26:26.29 It stops and starts every few hundred nucleotides in human cells,
00:26:32.22 and every time you want to start a new one, you have to make a new RNA primer
00:26:36.29 and that involves an enzyme, which is one of our 7 proteins,
00:26:41.01 called a DNA primase. It's a special protein that makes a short RNA primer,
00:26:46.24 a few nucleotides long, just long enough to start the synthesis by the DNA polymerase each time.
00:26:53.18 You can see the single-strand DNA binding protein coating all the single-stranded DNA.
00:26:59.00 It's probably also ahead of the DNA polymerase molecule on the lagging strand,
00:27:04.10 not indicated there. The new molecule I haven't described is again one of our seven proteins,
00:27:10.11 was an enzyme called a DNA helicase.
00:27:14.08 Very important enzyme which uses ATP energy, hydrolyzing ATP,
00:27:19.07 to propel itself along the DNA and opening up the helix
00:27:24.22 ahead of the leading strand DNA polymerase to make the DNA single-stranded
00:27:29.02 in the form that the DNA polymerase needs in order to copy that template.
00:27:34.01 Now, the fork is actually folded, this was our, probably, my most exciting discovery,
00:27:41.27 the realization, basically, I was giving a seminar somewhere
00:27:48.01 and I was, like I'm doing now, and I suddenly realized that it made no sense
00:27:54.25 for the DNA polymerase on the lagging strand to have to come on and off every time it made an Okazaki fragment,
00:28:00.29 that I could actually test whether, as a biochemist,
00:28:04.27 whether it was being retained, and used over and over again
00:28:07.24 and we actually found that was true.
00:28:09.17 So, the fork is actually folded to allow the DNA polymerase on the lagging strand
00:28:13.08 to be used over and over again,
00:28:15.17 therefore allowing very rapid DNA synthesis.
00:28:19.06 So, the, really, the whole point of my talk really is to introduce you to this movie,
00:28:26.11 my favorite movie.
00:28:28.21 It was not made by me, it was made at Cold Spring Harbor laboratory,
00:28:32.24 by Bruce Stillman and coworkers who work on human and yeast DNA replication.
00:28:38.29 It was part of the 50 year DNA celebration there, 2003.
00:28:43.03 There was a big party, I was there, and this movie was produced and distributed
00:28:48.04 for everybody to use, originally through the Cold Spring Harbor website.
00:28:52.25 And I'll let this movie play, it's narrated, as it's narrated in our textbook,
00:28:59.15 and you can find this movie on YouTube if you want, as well as on my talk.
00:29:04.17 So, this shows the replication fork in action and what I want you to get from this movie
00:29:13.11 is the general idea of how all of life works.
00:29:18.08 It's, life, I can talk to you only because life, the biochemistry of life,
00:29:24.03 the chemistry of life, is this complicated and elegant.
00:29:27.21 (Female voice) "Using computer animation, based on molecular research,

00:29:35.20 You're looking at an assembly line of miniature biochemical machines
00:29:42.15 that are pulling apart the DNA double helix and cranking out a copy of each strand.
00:29:47.20 The DNA to be copied enters the production line from bottom left.
00:29:55.03 The whirling blue molecular machine is called a helicase.
00:29:59.10 It spins the DNA as fast as a jet engine as it unwinds the double helix into two strands.
00:30:05.17 One strand is copied continuously and can be seen spooling off to the right.
00:30:13.22 Things are not so simple for the other strand because it must be copied backwards.
00:30:20.07 It is drawn out repeatedly in loops and copied one section at a time.
00:30:25.13 The end result is two new DNA molecules."
00:30:32.00 Well, that movie I find inspiring,
00:30:36.23 it really shows this mechanism that we have been working on for so many years,
00:30:45.04 me and many other laboratories.
00:30:47.22 And it's so satisfying to work on mysteries for decades
00:30:54.07 and finally solve mysteries, so that's why it's my favorite movie.
00:30:57.16 So, now, it turns out that almost every process in the cell
00:31:03.15 is recognized to be driven in a similar way,
00:31:06.20 different mechanisms, by a complex of ten or more proteins
00:31:10.21 that works like what we call a protein machine.
00:31:14.03 These protein machines function very much like the machines in everyday life
00:31:18.04 that are driven by electric energy, except that of course, they're not driven that way,
00:31:21.25 they also need ordered movements, in this case ordered movements of protein and RNA molecules
00:31:29.06 and those ordered movements are driven by proteins in the set that specifically hydrolyze a nucleotide, ATP or GTP,
00:31:36.17 to give it unidirectional motion and to harness the energy of that hydrolysis
00:31:44.24 to make the machine be effective, because you need ordered movements to make machines.
00:31:53.06 That's why we use electricity to drive machines in everyday life.
00:31:58.15 So, here are some, now at the end of my talk,
00:32:03.23 this is basically some personal lessons that I've learned as a scientist.
00:32:09.13 I guess the most important one is not listed here, which is that being a scientist is exciting,
00:32:16.03 you get to get paid for solving problems that both are fascinating to try to solve,
00:32:26.10 puzzles, but also, in the end, turn out to be useful,
00:32:29.22 because although we weren't intending this, the kind of information we discovered
00:32:34.01 is very important for our understanding of what happens in human diseases
00:32:39.08 where these things go wrong, including cancer.
00:32:42.04 But, the major mystery, the major point is, to work on something that's important that you don't understand.
00:32:50.25 I call them mysteries because if you then solve that problem,
00:32:54.27 it will help the whole world understand
00:32:58.22 whatever process your process is connected with much better.
00:33:05.21 I mean, obviously, we needed to know how DNA replicated,
00:33:09.10 that, we didn't know what it would be useful for,
00:33:11.00 but it obviously has helped us understand many other things.
00:33:15.10 Second thing which is, I've been stressing, and it often gets lost
00:33:20.15 in this push to, you know, to go take government grant money
00:33:25.08 and solve some disease problem, is that the shortest path
00:33:32.26 to actually solving those kinds of human problems often lies in backing up a bit
00:33:38.12 and coming to understand how basic processes work.
00:33:45.06 And this is often best done not in human cells, but in a model organism,
00:33:49.14 like the one I used, or a bacteria, or a yeast cell,
00:33:52.12 which are, all of which are much more readily manipulatable
00:33:56.23 and easier to understand than our human cells.
00:34:01.07 And finally, of course, the major lesson from my PhD thesis is that
00:34:06.09 in many ways the Watson and Crick discovery, which was so critical,
00:34:10.20 was unique. Most theoretical predictions of biologists turn out not to be correct
00:34:21.12 and to be oversimplified because nearly all cell processes will be based on elegant mechanisms
00:34:28.06 which are too hard to predict, we just have to go find out how it works by experiment.
00:34:34.11 So, thank you for listening, I hope this helped you to better understand a basic process that's going on
00:34:42.12 as, every minute of the day, in billions of cells in your body,
00:34:47.25 if it wasnâ€™t going on, of course, you'd be in very bad trouble, you couldn't be alive.
